Comments: Rb is inactivated in most glioma cells, and intact in normal cells. Delta-24RGD is an adenovirus that has been engineereed so that it cannot replicate in cells that contain Rb protein. Thus Delta-24 RGD replicates preferentially in cancer cells. In animal experiments, 60% of animals survived until they were euthanized, and microscopic examination of brains showed complete regression of tumor. No observable toxicity was found preclinically. Tumors must be either surgically accessible or accessible for stereotactic injection, and no more than 5 cm in diameter. Physicians must agree that treatment path will not deliver adenovirus to the ventricular system. Patients with Li-Fraumini Syndrome, germline deficit in retinoblastoma gene or related pathways, and immuno-compromised patients are not eligible.
