Comments: Suberoylanilide hydroxamic acid (SAHA) is a histone deacetylase inhibitor that has demonstrated potent antitumor effects in vitro and in vivo and shows significant synergistic activity against glioblastoma when combined with either temozolomide or BCNU. Early results from phase I trials suggest that SAHA is well tolerated, and dose-limiting toxicities do not overlap with those of temozolomide.
