In this study, lapatinib will be tested in children with brain tumors that are either recurrent (the disease has come back), or refractory (the disease was not responsive to therapy). In laboratory and animal research studies, lapatanib was able to block the receptor site on the cell (a site on the surface of the cell which acts as a lock and key), which signals a brain tumor cell to divide and grow. Lapatinib has been tested in adults and some evidence has shown that it can shrink, or at least stop the tumor from growing. We are now testing this new experimental drug in children, with the hope of finding a new drug that works against childhood brain tumors. This is the first time this drug will be tested in children.
Lapatinib will be given orally (in pill form), twice a day, for 28 days. Each 28 day period of time is called a cycle. A total of 26 cycles (two years) of the drug may be administered if there are no unacceptable side effects or tumor growth.
