Temsirolimus is a rapamycin ester which inhibits mTOR, and has clinical activity in glioblastoma multiforme, breast cancer and mantle cell lymphoma.[1][2][3]
Temsirolimus is a dihydroester of rapamycin that is given intravenously and is undergoing phase II testing in a variety of cancers.
Researchers from the Mayo Clinic treated 65 patients with recurrent glioblastoma multiforme. Non-hematologic toxicities included hypercholesterolemia, hyperglyceridemia and hyperclycemia. Neuroimaging revealed improvement in 36% of patients with responding patients having a longer survival than non-responding patients. They also observed that high levels of phosphorylated p70s6 kinase in baseline tumor samples appeared to predict response.
